If you can't leave that screen click the X at the top right hand side to close that window.

If the Test Now is flashing then you need to click on them so it will test the drives again. When you installed 4.09d that reset the drives to a not tested state.
If that does not fix it re-install Hoster and on the Personal Information page of the installation check the box at the bottom to clear the registry settings. Then open Hoster, retest the drives, set which drive you are going to use and Save.
Are you using an XP or Vista computer?

I click on the "Test Now" button but it seems kind of fade, so nothing works if I click there. The weird thing is that I have one drive which it detect as a good, and select that drive but the only thing I have is a small yellow ballon that said "untested drivers" "Please test all drivers before you living this dialog" and nothing else happen. I will try to reinstall it and see what happen.

ok I did the reinstallation, now all my drivers look green and good, but still the same yellow ballon that said "untested drivers" "Please test all drivers before you living this dialog" and can't cancel or save or do anything there.

What else should I do, any ideas?

I'm a bit lost for an answer at this point might be best to wait and see if anyone else has an idea.
One thing you could try, start Hoster34 which comes with all the 4.xxx series. It should be in folder that you downloaded Hoster to and see if that works then go back to 4.09d.

Is the operating system XP or Vista?
Are you logged in as an administrator?

If it is XP go to this link and make sure you have done all the steps:
http://www.mtu.com/upgrades/hoster/xp-changes.htm

If it is Vista then go to this link:
http://www.mtu.com/upgrades/hoster/v...de-hoster4.htm

After you do that do the Remove and Re-install Hoster. Be sure to check the box on the personal information page to clear the registry entries as it installs.
