I don't think you can change the default location but if you "Save as" you can select another folder to save to, it should then reopen using that location but pressing save or new will revert back to the default location.

You say "Carbonite doesn't want to back up my program data file" When you choose the option to backup, Hoster closes then the file path shown in the black window is set can't be altered that is the file you are backing up.
After you press the backup button that should take you to an explorer dialog where you should be able to select your Carbonite back up drive.
If for some reason you can't select that from explorer I suggest you save it to another location, it saves it as a zip file to a name of your choice, then come out of that dialog then move the saved file to your Carbonite cloud drive.
