The way I am reading this is that F: is an internal drive and G: is an external. If that is the case then no it won't work as you state.
You cannot have both in the Rebuild list at the same time. If you do you will get a duplicate for each song file you have. In addition the Rebuild uses all drive folders listed, so highlighting one or the other does not change how it works.
Basically, there is no fast way to switch from one location to the other.
To switch between the 2 drives you would have to remove the F: drive from the Rebuild list and add the G: drive to the list then rebuild. Depending on how many files you have the time to rebuild will increase the more you have.
If they are both external drives and the folder(s) for your song files are exact duplicates including the name of the folder then as Billyo said you should be able to swap out the drives without having to rebuild. This would have to be done while the computer is off and using the same USB plug, so that both drives come up as F:.
